Somatic embryogenesis and plant regeneration from gladiolus callus in uitro


Abstract
Somatic embryos were induced directly on cormel tissue explants or indirectly on callus of Gladiolus. Plantlets regenerated from the somatic embryos. External characteristics of embryogenic callus were white and compact, and organized. The ability of somatic embryogenesis was differed from cultivars, however, no difference with callus ages. Embryogenic capacity has been maintained for more then 28 months by subculturing callus at one month intervals. Somatic embryos were most efficiently formed on Murashige and Skoog basal medium containing either 0.075-0.1 §·/§¤ 2,4-D or 0.1-1.0 §·/§¤ NAA. Development of somatic embryos was poor in medium contained 2, 4-D, but favorable in medium added NAA . The early stage of somatic embryos were similar with zyfotic embryos and later developed into plantlets.
